General description
Nucleophosmin (UniProt P06748; also known as B23, NPM, Nucleolar phosphoprotein B23, Nucleolar protein NO38, Numatrin) is encoded by the NPM1 (also known as NPM) gene (Gene ID 4869) in human. Nucleophosmin (NPM) is a nucleolar protein that plays a role in ribosome biogenesis, nucleocytoplasmic transport, centrosome duplication, embryonic development, histone chaperone function, and transcriptional regulation. NPM is phosphorylated by Cdk2-cyclin E on Thr199 which causes the dissociation of NPM from the centrosome to allow duplication during the cell cycle. NPM is phosphorylated by the mitotic Cdk1-cyclin B complex on Thr234/237 during mitosis, resulting in its reassociation with the duplicated centrosomes. The human NPM1 gene transcribes three alternatrively spliced isoforms, isoforms 1&2 contain a nucleolar localization signal at their C-terminal end (W288/290 of isoform 1 and W259/261 of isoform 2), which is missing in the spliced isoform 3. In addition, a nuclear export signal is introduced, rendering the aberrant cytoplasmic location of this spliced isoform (NPMc).
